NAD+ (β-nicotinamide adenine dinucleotide, oxidized form) is a dinucleotide coenzyme present in essentially every living cell and one of the most extensively characterized molecules in biochemistry. It functions as the primary electron acceptor in catabolic metabolism and as a consumed substrate for an entire class of signaling enzymes.

Research interest divides along those two roles. As a redox cofactor, NAD+ is central to glycolysis, the citric acid cycle, and oxidative phosphorylation. As a substrate, it is consumed by sirtuins, PARPs, and CD38 — which means cellular NAD+ availability directly constrains DNA repair, deacetylation signaling, and immune enzyme activity.

Solas Science supplies NAD+ as a research-grade preparation in a metered nasal spray format at a labeled 10 mg per spray, prepared under controlled conditions with quality procedures designed to support consistency, reliability, and analytical reproducibility.

For laboratory research use only. Not for human or veterinary use. Not a dietary supplement, food, drug, or cosmetic. Not for diagnostic or therapeutic application.

What Is NAD+?

NAD+ (CAS 53-84-9) has the molecular formula C21H27N7O14P2 and a molecular weight of 663.43 g/mol. It consists of two nucleotides — one carrying nicotinamide, the other adenine — joined through a pyrophosphate bridge.

The nicotinamide ring is the functional center. It accepts a hydride ion to become NADH and releases it to return to NAD+, and that reversible two-electron transfer is the basis of hundreds of characterized enzymatic reactions. The NAD+/NADH ratio, rather than the absolute concentration of either, is what most redox research actually measures.

The second role is distinct and non-catalytic: sirtuins, poly(ADP-ribose) polymerases, and CD38 cleave NAD+ and consume it outright. Because the pool is finite, competition between these consumers is itself an active research subject, and is the reason NAD+ availability appears so often in work on DNA damage response and metabolic signaling.

About the Metered Nasal Spray Format

This preparation supplies NAD+ as an aqueous solution in a 10 mL bottle fitted with a metered pump delivering a labeled 10 mg per actuation — approximately 100 metered sprays per bottle, or 1,000 mg of compound in total. Actual spray count varies with pump priming and residual volume.

NAD+ is the outlier in this format: it is dosed in milligrams rather than micrograms because it functions stoichiometrically as a coenzyme rather than catalytically at a receptor, so useful experimental concentrations are orders of magnitude higher than for the peptides above. It is also the least stable of the five in aqueous solution, which makes refrigeration and a recorded open-date more consequential here than elsewhere.

Researchers who need to control concentration themselves, or who are working in cell culture and solution-phase assays, may prefer the lyophilized powder presentation, which is reconstituted to a concentration of the investigator’s choosing. This metered format trades that flexibility for a fixed, pre-measured dose delivered without reconstitution.

Scientific Research Areas of NAD+

NAD+ is investigated across metabolic biochemistry, mitochondrial biology, DNA repair, and enzymology.

Redox Biology and the NAD+/NADH Ratio

The foundational research application concerns cellular redox state. Investigators examine:

  • NAD+/NADH ratio measurement in cytosolic and mitochondrial compartments
  • Dehydrogenase enzyme kinetics and cofactor dependence
  • Glycolytic and citric acid cycle flux
  • Redox-coupled metabolite profiling

Mitochondrial Metabolism and Bioenergetics

Mitochondrial research uses NAD+ to study:

  • Complex I substrate supply and electron entry into the respiratory chain
  • Oxidative phosphorylation efficiency and ATP yield
  • Mitochondrial NAD+ transport and compartmentalization
  • Fatty acid oxidation and substrate switching

Sirtuin and DNA Repair Signaling

The NAD+-consuming enzyme families are a major research focus:

  • Sirtuin deacetylase activity and substrate acetylation state
  • PARP activation following experimental DNA damage
  • CD38 activity and NAD+ pool depletion
  • Competition among NAD+-consuming enzymes for a shared pool

Biosynthesis, Salvage and Precursor Research

Work on how cells maintain the pool examines:

  • Salvage pathway flux via NAMPT and nicotinamide recycling
  • De novo synthesis from tryptophan
  • Comparison of NAD+ with precursor compounds such as NMN and NR
  • Age- and stress-associated changes in cellular NAD+ content

Handling and Storage

Store refrigerated at 2–8 °C, protected from light, and do not freeze — freeze-thaw cycling can degrade peptides in solution and damage the metering pump. Keep the bottle upright with the cap fitted, and clearly labeled for laboratory inventory. As an aqueous preparation this product has a shorter working life than a lyophilized powder; record the date the bottle is first opened and handle it accordingly.

Handle in accordance with standard laboratory safety practice, including appropriate personal protective equipment. The metering pump is intended for controlled dispensing in a laboratory setting only. Keep out of reach of children and unauthorized personnel.
